It goes in the story that their mother was murdered by something “supernatural.” After that occurrence, the 2 brothers spent most of their off-time trying to hunt this person and rid the world of supernatural evils. Go forward about 22 years and it shows Sam and Dean’s father being murdered by that same demon who took their mother.
